ABSTRACT

A retrospective case review was conducted of 3 cases with umbilical venous catheterization(UVC) related pericardial effusions in the Neonatal Intensive Care Unit of Zhongnan Hospital of Wuhan University from December 2020 to April 2022.All 3 cases were preterm infants with gestational ages of 33 + 4, 31 and 27 + 6 weeks, respectively.UVC was inserted routinely in 24 hours after birth.Three neonates developed tachycardia or bradycardia, dyspnea, decreased oxygen saturation and muffled heart sound at the 1 st to 4 th day after catheterization.Echocardiography indicated pericardial effusion, so the 3 neonates underwent pericardiocentesis and drainage.Among the 3 neonates, 2 cases improved and have good prognosis, 1 case died.UVC can cause pericardial effusion, which occurs mostly in the early stage after catheterization.Pericardial effusion and tamponade should be considered when patients show unexplained sudden clinical deterioration after catheterization, such as dyspnea, cyanosis, tachycardia or bradycardia, etc.Once diagnosed, umbilical vein catheter should be removed in time and pericardiocentesis and drainage should be performed for decompression.Early diagnosis and intervention can effectively improve the prognosis.

ABSTRACT

Objective: To establish a high performance liquid chromatography method for the determination of 2-thioxothiazolidine-4-carboxylic acid (TTCA) in urine. Methods: After acidification with hydrochloric acid, TTCA in urine was first extracted by ethyl acetate with excessive sodium chloride, then gradient separated by a symmetry C18 column and then detected by a diode array detector. The quantification was based on a working curve of external standard method. Results: The linear relationship of TTCA in urine was good in the range of 0.03-10.00 mg/L, and the correlation coefficient was 0.9999. The detection limit and minimum quantitative concentration of TTCA in urine were 0.008 mg/L and 0.027 mg/L. The intra-assay precision of the method was 0.9%-1.4%, the inter-assay precision was 1.3%-3.5%, and the average recovery was 85.0%-92.7% while the concentrations of TTCA in urine was 0.8, 2.0 and 8.0 mg/L, respectively (n=6) . Conclusion: The gradient elution high performance liquid chromatography method has simple operation and high sensitivity, and it is suitable for the determination of TTCA on a low level in urine for occupational workers exposure to carbon disulfide.

ABSTRACT

Objective:To observe the effects of modified Liuwei Dihuangtang on serum fibroblast growth factor 23 (FGF23), full-length intact parathyroid hormone (iPTH), and 1,25-dihydroxyvitamin D<sub>3 </sub>[1,25(OH)<sub>2</sub>D<sub>3</sub>] levels and Klotho and FGF23 protein expression in renal and bone tissues of rats exposed to high phosphorus combined with adenine, so as to explore the mechanism of modified Liuwei Dihuangtang against renal osteopathy. Method:One hundred and thirty healthy adult SD rats were randomly divided into five groups, namely normal group(<italic>n</italic>=10),high phosphorus group(<italic>n</italic>=30),model group(<italic>n</italic>=30),modified Liuwei Dihuangtang group(<italic>n</italic>=30) , and calcitriol group(<italic>n</italic>=30),and rats in each group were further classified based on three time points, namely 8,10, and 12 weeks. Rats in the normal group were fed with normal diet, the ones in the high phosphorus group with high phosphorus diet, and those in the other groups with adenine and high phosphorus diet for inducing renal osteopathy. Rats in the normal group,high phosphorus group, and model group were intragastrically administered with distilled water (10 mL·kg<sup>-1</sup>·d<sup>-1</sup>),the ones in the modified Liuwei Dihuangtang group with modified Liuwei Dihuangtang (2.556 g·kg<sup>-1</sup>·d<sup>-1</sup>) , and those in the calcitriol group with calcitriol (0.09 μg·kg<sup>-1</sup>·d<sup>-1</sup>). Result:Compared with the normal group and high phosphorus group at the weeks of 8,10 and 12,the model group displayed significantly elevated blood urea nitrogen(BUN),serum creatinine(SCr),serum phosphorus,iPTH,FGF23,renal interstitial fibrosis score, and FGF23 expression in renal and bone tissues, but lowered serum calcium and 1,25(OH)<sub>2</sub>D<sub>3</sub> and Klotho protein expression in renal and bone tissues(<italic>P</italic><0.05 ,<italic>P</italic><0.01). Compared with the model group at the weeks of 8,10 and 12, the modified Liuwei Dihuangtang and calcitriol both significantly decreased the serum BUN,SCr,serum phosphorus,iPTH, FGF23, tubulointerstitial semi-quantitative score, and FGF23 expression in renal and bone tissues, while increased the serum calcium,1,25(OH)<sub>2</sub>D<sub>3</sub>, and Klotho protein expression in renal and bone tissues (<italic>P</italic><0.05,<italic>P</italic><0.01). There was no significant difference in the above-mentioned indexes between the modified Liuwei Dihuangtang group and the calcitriol group at the same time point. Conclusion:Klotho-FGF23 axis is probably involved in renal osteopathy. The modified Liuwei Dihuangtang effectively improves renal function,alleviates pathological changes in renal and bone tissues,and regulates calcium and phosphorus metabolism to protect the bone, which is related to its regulation of Klotho-FGF23 axis.

ABSTRACT

Objective To disclose the molecular mechanism of calycosin-7-O-β-D-glucoside (CG) accumulation in Astragalus membranaceus, we cloned PAL genes and analyzed the expression patterns of them and changes of CG contents in different tissues of A. membranaceus. Methods PAL genes were cloned with the methods of homology cloning and RACE technique using the total RNA as template and the analysis of bioinformatics on the cloned genes was carried out, gene expressions in root, stem, and leaf were determined with real-time PCR method, and CG content in root, stem, and leaf were analyzed by HPLC methods. Results Three PAL genes were cloned from A. membranaceus. The genbank accession number was KY086279 (AmPAL1), KY086280 (AmPAL2), and KY086281 (AmPAL3), respectively; The full-length cDNA of them was 2 508 bp, 2 401 bp, and 2 498 bp, respectively; And they all consisted of 2 157 bp open reading frame encoding 718 amino acids. Deduced AmPAL proteins had typical active sequences of PAL proteins, they were homology with other PAL proteins, and they shared the highest identities with PAL proteins of leguminous plants. Phylogenetic tree analysis showed AmPAL1 belonged to the different sub-class with the sub-class of AmPAL2 and AmPAL3. Real-time PCR analysis indicated that expression levels of AmPALs were different from each other, the expression level of AmPAL1 was the highest, the expression level of AmPAL2 was the next, and that of AmPAL3 was lowest in all detected tissues, and only the expression levels of AmPAL2 was similar to the changes of CG contents in different tissues (root > stem > leaf). Conclusion The cloned AmPAL1, AmPAL2, and AmPAL3 from A. membranaceus were typical genes of PAL, each might have different function in developing of different tissues, and AmPAL2 might involve in CG accumulation in different tissues.

ABSTRACT

OBJECTIVE: To discuss the clinical efficacy and safety of balloon-expandable stent in the treatment of symptomatic atherosclerotic cerebral artery stenosis in the middle cerebral artery(MCA), and to assess the appropriate methods of postoperative follow-up auxiliary examination. METHODS: A total of 67 patients with symptomatic atherosclerotic cerebral artery stenosis in the MCA were treated with balloon-expandable stent. The clinical data and follow-up data were collected and then analyzed retrospectively.RESULTS: the success rate of the surgical treatment with balloon-expandable stent was 98.51%(66/67). The incidence of symptomatic perioperative complications was 4.48%(3/67). All patients received clinical follow-up, and the auxiliary examination methods of the internal condition lesion of the stent included transcranial Doppler(TCD) or digital subtraction angiography(DSA). The incidence of in-stent restenosis or occlusion was 8.96%(6/67), and the consistency rate of stent restenosis or occlusion between TCD and DSA was75.00%, and the incidence of symptomatic in-stent restenosis or occlusion was 2.99%(2/67). It was found that there were two patients with recurrent stroke in cerebrovascular stent related region but without in-stent restenosis, and within 1 year of follow-up, the incidence of symptomatic stroke in stent-associated areas after operation was 10.44%(7/67). CONCLUSION: The treatment with balloonexpandable stent for symptomatic MCA stenosis has good clinical effect. The incidence of perioperative complications is low and the incidence of stroke in stent-associated areas after operation is low. TCD is an effective way of follow-up after the arterial stenting in the brain, which can play a role in the early detection of the in-stent restenosis, and its consistency with DSA is high.

ABSTRACT

Janus kinase-signal transducers and activators of transcription (JAK-STAT) signal transduction pathway is related to cellular biological activities and occurrence and development of many diseases. In recent years, JAK-STAT signal transduction pathway has been found in the central nervous system to regulate neurodegenerative diseases and nerve regeneration after nerve injury: however, promoting endogenous neurogenesis as a new direction of nerve regeneration research is also closely related to the positive or negative regulation of JAK-STAT signal transduction pathway. This article reviews the research progress of JAK-STAT signal transduction pathway, neurogenesis, and JAK-STAT signal transduction pathway regulating neurogenesis.

ABSTRACT

Objective To evaluate the feasibility and safety of self designed oral-nasal double channel mask in painless gastroscopy. Methods 400 patients who were given intravenous injection Sublimaze followed by Propofol were divided equally at random into two groups. The patients in control group (n = 200) were inhaled oxygen with snuffle tube (4.0 ~ 5.0 L/min) and the patients in experimental group (n = 200) were inhaled oxygen with a self designed oral-nasal double channel mask. Respiration rate, SpO2, duration of operation and dosage of anaesthetic were recorded before and during operation. Result The SpO2decreased markedly and duration of operation obviously prolonged in group I during operation (P < 0.05), there was no statistical difference between the two groups of the respiration rate and dosage of anaesthetic. Conclusion The application of oxygen inhalation with the self designed oral-nasal double channel mask in painless gastroscopy appeared safer than that of oxygen inhalation via snuffle tube. It can be safely used in painless gastroscopy.

ABSTRACT

<p><b>OBJECTIVE</b>To explore whether fractional anisotropy (FA) value could be taken as a quantitative indicator for tracing and reexamining amyotrophic lateral sclerosis (ALS), and to analyze the correlation between FA value and integrative medical treatment.</p><p><b>METHODS</b>Totally 18 ALS patients were recruited in this study. All patients received diffusion tensor imaging (DTI) using 3. OT (Propeller HD) MRI twice. Six regions of interest (ROI) were selected to measure FA values. Survival analyses were performed in 11 cases of end point events.</p><p><b>RESULTS</b>(1) Three ROI (cerebral peduncle, posterior limb of internal capsule, and corona radiata) all indicated that FA value was the highest in patients with mild health status scale of amyotrophic lateral sclerosis (ALS/HSS). (2) There was statistical difference in the means of FA values in cerebral peduncle, posterior limb of internal capsule, and corona radiata of 18 cases between initial examination and reexamination (P < 0.01). (3) Kaplan-Meier survival curve showed the survival rate of ALS patients decreased as time went by, with the median survival time of 48 months.</p><p><b>CONCLUSIONS</b>FA value was inversely proportional to the severity of ALS, the more severe, the lower FA values. FA value was an objective indicator for assessing the severity of ALS. ALS is an incurable disease till now. Integrative medical treatment might become one direction for ALS patients.</p>

ABSTRACT

<p><b>Objective</b>To construct a GFP-fused mouse Parkin co-regulated gene (PACRG) baculovirus recombinant PACRG/GFP-pFastBac1 vector and express the fusion protein in Sf9 insect cells.</p><p><b>METHODS</b>Full-length mouse PACRG cDNA was amplified by PCR and cloned in frame to the vector pFastBac1 with eGFP (rpFBac-PACRG-GFP recombinant vector). The plasmid was transformed into DH10Bac cells to obtain the recombinant bacmid plasmid, the bacmid was transfected into Sf9 insect cells, and the expressed PACRG/GFP fusion protein was analyzed by Western blot and fluorescence microscopy.</p><p><b>RESULTS</b>The construction of the PACRG/GFP-pFastBac1 baculovirus plasmid was confirmed by sequencing and restriction enzyme digestion. Western blot showed the expression of the fusion protein carrying a green fluorescence in the Sf9 insect cells.</p><p><b>CONCLUSIONS</b>Conclusion: A PACRG/GFP-pFastBac1 recombinant baculovirus vector was successfully constructed and the fusion protein was highly expressed in the Sf9 insect cells. Our findings have provided a basis for further studies on the structure of the PACRG protein and regulation of spermatogenesis.</p>

ABSTRACT

<p><b>Objective</b>To construct eukaryotic expression plasmids of the Tac1 gene and explore the interaction between TAC1 and sperm-associated antigen 6 (SPAG6).</p><p><b>METHODS</b>RNA was extracted from the heart, liver, spleen, lung, kidney, brain, muscle, and testis of 10 Kunming male mice and, after reverse transcription into cDNA, the expression of Tac1 in the above tissues was observed by RT-PCR. Tac1/pEGFP-N2 and Tac1/pGADT7 recombinant plasmids were constructed and Tac1/pEGFP-N2 was transfected into CHO and COS-1 cells, followed by localization and detection of the protein expression of TAC1 by immunofluorescence staining and Western blot. The interaction between TAC1 and SPAG6 was determined by yeast two-hybrid experiment and Western blot.</p><p><b>RESULTS</b>Tac1 was expressed mainly in the testis, brain and heart. The results of restriction enzyme digestion and sequencing indicated successful construction of the recombinant plasmids, with the restriction fragment length of 390 bp. TAC1 was localized in the whole body of the CHO cells when transfected alone, but expressed in the microtubule of the cells when cotransfected with SPAG6, with the molecular weight of 40 000. Yeast two-hybrid experiment showed the colonies of TAC1 and SPAG6 on the culture plate without Leu, Trp and His, both contained in the yeast fusion protein.</p><p><b>CONCLUSIONS</b>The Tac1 recombinant plasmid was constructed successfully and the interaction between TAC1 and SPAG6 was confirmed with the plasmid.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the frequency distribution of single nucleotide polymorphisms (SNPs) in two genes associated with incomplete Kawasaki disease (KD) (rs1569723 in CD40 gene and rs2736340 in BLK gene), and to investigate its association with the genetic susceptibility and clinical phenotypes of incomplete KD.</p><p><b>METHODS</b>A total of 184 children with incomplete KD and 203 normal children were recruited to carry out a case-control study. The genotypes of SNPs in CD40 gene and BLK gene were determined using polymerase chain reaction-restriction fragment length polymorphism. The frequency distribution of genotypes was compared between the KD and control groups. The association between gene polymorphisms and clinical features of incomplete KD was analyzed.</p><p><b>RESULTS</b>There were no significant differences in genotype (AA, AC, CC) and allele frequencies in CD40 SNP rs1569723 between the KD and control groups. There were significant differences in the frequency distribution of three genotypes (TT, CT, CC) in BLK SNP rs2736340 between the KD and control groups (P=0.031), and the KD group had a significantly higher frequency of T allele than the control group (P=0.007). There were significant differences in the incidence of conjunctival hyperaemia among the patients with different genotypes (rs1569723 in CD40 gene) (P=0.036). The SNP rs2736340 in BLK gene was associated with the extremity changes in KD patients (P=0.017).</p><p><b>CONCLUSIONS</b>The SNP rs2736340 in BLK gene is associated with the susceptibility to incomplete KD, and the SNP rs1569723 in CD40 gene and SNP rs2736340 in BLK gene are associated with some of clinical phenotypes of incomplete KD.</p>

ABSTRACT

Objective To investigate myocardial apoptosis in the septic rats and to evaluate the role of Bcl-2, Caspase-3 in signal transduction of myocardial apoptosis. Methods Sixty Wistar rats were randomly divided into control group (n=30) and septic group (n=30). Either group was then divided into 5 subgroups:3 h,6 h,12 h,24 h and 36 h subgroup. Sepsis model was established by cecal ligation and puncture (CLP). Control group were given the same treatments except CLP. TUNEL method was used to determine apoptosis index (AI). Western blot was applied to determine Bcl-2 and Caspase-3 expression. Results (1) AI value increased progressively after CLP procedure and peaked at 36 h(43.742±2.495), which showed significant difference compared with control group(4.311±0.035 ). There was no significant difference between 24 h and 36 h subgroup (P>0.05). (2) Bcl-2 protein levels significantly decreased after CLP procedure compared with control group (P<0.01 ) and 36 h subgroup was the lowest, but there was no significant difference between 24 h and 36 h subgroup (P>0.05). Caspase-3 levels gradually increased after CLP procedure,and 36 h subgroup was the highest,but there was no significant difference between 24 h and 36 h subgroup ( P>0.05). (3) There was negative correlation between Bcl-2 and AI (r=-0.753, P<0.001). Conclusion Myocardial apoptosis may be one of mechanisms for myocardial injury in sepsis. Anti-apoptosis intervention may be a promising therapy to improve the prognosis of sepsis.

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the expression level of candidate tumor suppressor gene N-Myc downstream-regulated gene 2 (NDRG2) in human colon cancer.</p><p><b>METHODS</b>Thirty samples of colon cancer tissues with matched normal colon tissues were collected. The NDRG2 mRNA level was detected by semi-quantitive RT-PCR and the NDRG2 protein level was examined by Western blot and immunohistochemistry.</p><p><b>RESULTS</b>Twelve samples of colon cancer tissues had low NDRG2 mRNA level and low protein level. The positive rates of NDRG2 in normal tissues and the tumorous colon tissues were 90.0%(27/30) and 53.3%(16/30) by immunohistochemistry respectively. There was a significant difference between two groups (P<0.05). The NDRG2 expression was not correlated with age, sex, metastasis of lymph node, depth of infiltration, as well as the Dukes staging(P>0.05), while it was correlated to the histology grading. The positive rate of NDRG2 in the well- and moderate-differentiation group was higher than that in the poor-differentiation group(P<0.05).</p><p><b>CONCLUSION</b>The expression of NDRG2 is low in some colon cancer tissues, which indicates that the low level of NDRG2 expression may be engaged in the development of colon cancer.</p>

ABSTRACT

<p><b>AIM</b>To study the regulation mechanism of GP on plasma lipoprotein metabolism and To explore its mechanism of anti-lipoperoxidation in the experimental hyperglycemia rats.</p><p><b>METHODS</b>The rats were raised with high fat diet for six weeks,and the model of hyperglycemia was then established. After that, those rats were treated with high or low dose of GP and xuezhikang as a masculine comparison for four weeks. Then, those rat were executed, and detected the plasma TC, TG, LDL-C, HDL-C, GSH-Px, at the same time the SOD, CAT and MDA concentration also be mensurated.</p><p><b>RESULTS</b>The results showed that high and low dose of GP could decrease the concentration of serum LDL-C, cholesterol and triglyceride remarkably and raise the concentration of HDL-C. The activity of GSH-Px, SOD and CAT in GP groups were promoted and the level of MDA was decreased distinctly.</p><p><b>CONCLUSION</b>The GP can therapy the abnormity of serum lipid and has obviously anti-lipoperoxidation affection.</p>

@#ObjectiveTo observe the effect of mild hypothermia on vital signs of the elderly patients with anterior circulation brain infarction (massive hemispheric infarction) and hyperpyrexia.Methods46 elderly patients with massive hemispheric infarction and hyperpyrexia within the first 24 hours after onset were randomly divided into the control group and the hypothermia group with 23 cases in each group. The patients of the hypothermia group were treated with mild hypothermia therapy (33℃~35℃). Those of the control group were treated with routine hypothermal treatment. The temperature and heart rate of all patients were measured after 10 days; the prognosis was evaluated according to mortality and scores of Neurological Deficit Scale (NDS).ResultsFor patients of the hypothermia group, the temperature and heart rate significantly reduced, death rate was significantly lower, the neurological function distinctly improved and prognosis was better than the control group after 10 days treatment ( P<0.05).Conclusion Mild hypothermia is safe and effective to elderly patients with massive hemispheric infarction and hyperpyrexia.

Objective To study the effect of Yiqimingmu pill on ATPase and LD of retinal ischemia rabbit, and probe into its mechanism. Methods Thirty-two Japanese large-ear white rabbits were divided into four groups, normal group (A group), model group (B group), comparison group (C group) and treated group (D group). Retinal ischemia was made in B, C and D group. After the treatment of one month, testing the correspondent indexes and studying with statistics. Results After the treatment of one month, in B and C group, the content of ATPse was lower, while the content of LD was higher compared with A group. Conclusion Yiqimingmu pill is reliable in the treatment of retinal ischemia, and superior to Buzhongyiqi wan.

<p><b>OBJECTIVE</b>To detect the function of proteolipid protein (PLP) peptide (residues 56 - 70)-specific CD(4)(+) T cells in experimental allergic encephalomyelitis (EAE) in Biozzi AB/H mice (H-2A(g7)).</p><p><b>METHODS</b>Biozzi AB/H mice were immunized by synthetic PLP(56 - 70) peptide (DYEYLINVIHAFQYV) which was emulsified by sonication with complete Freund's adjuvant, a EAE model proven histologically and clinically. Murine splenocytes and spinal cord infiltrated (SCI) T cells were stimulated by PLP(56 - 70), then the CD(4)(+) T cells were isolated by Dynabeads, and confirmed by staining with anti-CD(4) antibody. Finally, the IL2 bioassay and IFN-gamma/IL4 ELISA were done to detect T cell proliferation and cytokine secretion after PLP(56 - 70) stimulation.</p><p><b>RESULTS</b>The histology of murine spinal cord showed a great number of lymphocytes infiltrated the spinal cord; the clinical signs showed high scores (4.3) on the peak, as well as a good EAE model. After being isolated by Dynabeads, CD(4)(+) T cells showed high purification (> 99%) by staining with anti-CD(4) antibody. IL2 bioassay showed that those T cells were PLP(56 - 70)-specific T cells. ELISA showed that those T cells had high IFN-gamma/IL4 ratio, indicating that they are T helper 1 (Th1) cells.</p><p><b>CONCLUSIONS</b>PLP(56 - 70)-specific splenocytes and SCI CD(4)(+) T cells in EAE from Biozzi AB/H mice were detected and showed that both of them were PLP(56 - 70)-specific Th1 cells. It is beneficial to understand what kind of role these T cells play in the development of EAE.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the cross-reactivity between glutamic acid decarboxylase (GAD)-I-A(g7) and I-A(d) tetramer in diabetes-prone non-obese diabetic (NOD) mice (I-A(g7)) and diabetes-free Balb/c mice (I-A(d)).</p><p><b>METHODS</b>Two GAD peptide I-A(g7) and I-A(d) tetramers were generated and compared for phenotype and function of sorted GAD peptide I-A(g7) and I-A(d) tetramer-positive (tet+) T cells.</p><p><b>RESULTS</b>The cross-reactivity is shown in either tetramer positive percentage or tetramer staining intensity. The NOD and Balb/c derived-tet+ T cells were able to be cross-stained by GAD peptide I-A(g7) and I-A(d) tetramers, and responded to both irradiated NOD and Balb/c splenotyes under stimulation by synthetic and recombinant GAD peptides.</p><p><b>CONCLUSION</b>Although I-A(g7) and I-A(d) are closely related in biochemical and biological aspects, their most notable difference is the presence or absence of a negatively charged residue at position beta57 that links to insulin-dependent diabetes mellitus.</p>

ABSTRACT

AIM: To establish an analytical method for detecting 14 kinds of amino acid in Wuling Capsule(xylaria) by HPLC-FLD. METHODS: After being derivated in precolumn with o-phthalaldehyde and ?-mercaptoethanol,the amino acids were eluted by gradient elution and were detected with a fluorescence detector (Ex = 340 nm,Em = 450 nm). The mobile phase consisted of 0. 1 mol/L NaH2PO4 ( adjusted to pH 6. 8 by phosphoric acid) and methanol with a flow rate of 1. 0 mL/min,column temperature was set at 30 ℃. RESULTS: The deriva-tives of 14 amino acids with o-phthalaldehyde could be separated with a stable base line within 40 min. The linear range of the detection was 5-200 nmol/mL and correlation coefficients were over 0. 999 0. CONCLUSION: The method is simple,accurate and well reproduced for the determination of amino acids in Wuling Capsule.
